Dissolution method development requires careful selection of an appropriate dissolution medium, generally aqueous buffer systems spanning the physiological pH range, chosen to reflect the solubility characteristics of the drug and, where relevant, to achieve sink conditions in which the dissolution medium volume is sufficient to maintain drug concentration well below saturation solubility throughout the test. Apparatus selection, rotation speed, medium volume, and sampling time points are optimised to produce a discriminating dissolution profile capable of distinguishing between batches of differing quality while remaining sufficiently robust to yield reproducible results across laboratories. Surfactants may be incorporated into the dissolution medium to enhance the solubility of poorly water-soluble drugs and thereby maintain sink conditions.
Practical Considerations and Critical Parameters
Selection of an appropriately discriminating dissolution method requires a careful balance, since a method that is too discriminating may generate excessive batch-to-batch variability unrelated to true product quality, whereas a method that is insufficiently discriminating may fail to detect meaningful differences in formulation or manufacturing performance.
